TD LIPO STUDY
A recent study conducted by University of Victoria's Laboratory for Innovations in Microengineering investigated the effects of three base creams, TD Lipo, an industry leading liposomal base, and your typical PLO-based gel, on the penetration and retention of a probe (fluorescein-free acid, FFA) through porcine skin. The results showed that TD Lipo enhanced both penetration and retention of FFA compared to other base formulations.
The study found that TD Lipo showed significantly higher skin penetration of the FFA than typical PLO-based gel and typical liposomal base at all sampling times. The difference in skin penetration between TD Lipo and 80% DMSO (a positive control) increased as sampling times went by. TD Lipo also showed significantly higher skin retention of the FFA than other base formulations.Â
Skin Retention and Penetration Performance
The Skin Retention Test highlights TD Lipo's exceptional ability to deliver and retain active ingredients within the skin. This test measured the amount of fluorescein-free acid (FFA) absorbed into the skin layers using a tape-stripping technique. By progressively removing the stratum corneum (SC) in 12 layers, researchers quantified FFA retention at different depths within the skin.
The graph illustrates the cumulative penetration profile of FFA across various formulations. TD Lipo consistently outperformed typical PLO-based gels and liposomal bases, achieving significantly greater penetration and retention over time. Unlike the other bases, TD Lipo exhibited a clear concentration gradient, with high retention near the application site and effective transport through deeper layers of the SC. This superior performance ensures more effective, sustained delivery of active ingredients, making TD Lipo a standout choice for transdermal drug delivery.
Visualization of Skin Penetration with Fluorescent Microscopy
The Microscopic Imaging Test used advanced fluorescent microscopy to visualize the depth and distribution of fluorescein-free acid (FFA) within the skin after treatment with TD Lipo. Cross-sectional slices of treated skin were prepared, and high-resolution images were captured to observe FFA penetration and retention across different formulations.
The graph and images reveal that TD Lipo achieved a higher and more uniform concentration gradient compared to typical PLO-based gels and liposomal bases. The fluorescence intensity near the application site was significantly higher with TD Lipo, confirming enhanced retention, while the gradient decreased predictably deeper into the skin layers. This uniform distribution pattern indicates that TD Lipo not only delivers active ingredients effectively but also ensures sustained absorption into the skin, making it an advanced solution for transdermal applications.
